Changes of arachidonic acid metabolism of M. leprae infected macrophages have been studied. Elevated level of PGE2 has been confined in supernatant of cultured macmphages from human lepromatous leprosy granuloma, lepromin A injected human monocytes, and murine peritoneal macrophages in this laboratory. There was no report regarding PGE2 level and superoxide anion(O2) production of M. leprae infected human coloshal macrophages(CM), which can play some roles in the infection route of leprosy in the neonate. CM will be the source of M. leprae infection into the neonate. This study was conducted to evaluate PGE2 level and superoxide anion production of M. leprae infected CM. The results are as follow : 1. CM Produced PGE2, 1,130¡¾ 80pg/§¢, about three times more than that of RPMI-human AB serum, but activation of CM with interferon-¥ã(IFN-¥ã) suppressed level of PGE2significantly(P<0.01). 2. Indomethacin blocked PGE2 production of both control and IFN-¥ã activated CM to the baseline. 3. Two doses of PGE2 blocked superoxide anion production of CM, althouh IFN-¥ã treatment enhanced superoxide anion production. 4. M. leprae treatment(1 : 2 and 1 : 20) suppressed production of superoxide anion of CM. Contrary to this, PMA(phorbol myristate acetate) and IFN-¥ã enhanced superoxide anion in kinetic study(P<0.01). |
