Two staining methods, the carbol fuchsin with acetic acid differentiation and the periodic acid-carbol pararosanaline, were used for demonstrating leprosy bacilli in skin smears of leprosy patients. The skin smears from 308 long-treated patients with lepromatous, borderline and tuberculoid leprosy were stained with periodic acid-carbol pararosaniline. There were no significant differences in BI and MI determinations than with classic carbol fuchsin staining, however slightly higher BI and MI results were observed than with latter staining. With the former stain, leprosy bacilli were found in 8 of 234 skin smears in which no bacilli could be seen by the batter stain. It is considered that under the action of antileprosy drugs some leprosy bacilli may lose their acid-fastness and become chromophobic and these chromophobic bacilli can be restored their acid-fastness in their staining with periodic acid pretreatment as reported by Dr. Harada etc. |
